off the top of my head, and purposely without picking more than one character from any show.....
1. Special Agent Dale Cooper - "Twin Peaks"
2. Edina Monsoon - "Absolutely Fabulous"
3. Jerri Blank - "Strangers With Candy"
4. Barnabas Collins - "Dark Shadows"
5. John Crichton - "Farscape"
and since picking only 5 was just too hard....
6. Basil Fawlty - "Fawlty Towers"
7. Rupert Giles - "Buffy the Vampire Slayer"
8. Ruth Fisher - "Six Feet Under"
9. Delenn - "Babylon 5"
10. Captain Malcolm Reynolds - "Firefly"

I posted this before reading any other entries, so as not to be "tainted." These are in alphabetical order, as I couldn't bear to "rank" them.
Reverend Jim Ignatowski ("Taxi")
Vic Mackey ("The Shield")
Lucy Ricardo ("I Love Lucy")
Homer Simpson ("The Simpsons")
Christian Troy ("Nip/Tuck")

Herman Munster-The Munsters
Theo Kojak-Kojak

Gotta throw out some love for (eschewing animated series):
Jack Bauer (24)
Jack Bristow (Alias)
Data (Star Trek: TNG)
Cosmo Kramer (Seinfeld)
The Lone Gunmen (The X-Files)
Is including the Lone Gunmen cheating?

He's #1 no question in my book. It is just too hard to limit myself to 4 others.
