I've never laughed as much or as hard at any movie as I did with Raising Arizona.

If you're at all well-read, Woody Allen's Love and Death is the funniest movie of all time.

Super Troopers is good, but it lags in a lot of scenes.

From the last couple of years, I would say Zoolander, Harold and Kumar, and Old School. In the 90's, I'd say Billy Madison, Mallrats, and either Tommy Boy or Black Sheep (same movie).

One of my personal favorites is Let It Ride, but that's just me.

I think Airplane and the Naked Gun movies are brilliant. And I have yet to laugh at a single Mel Brooks film, and this is after watching Young Frankenstein, that Hitchcock/psychiatric movie, Spy Hard, and Spaceballs. I might have smiled, but I have never laughed. *shrug*

I'm not much into slapstick or infantile poopy humor (gotta be in the right mood) but my two favorite (semi-intelligent) comedies of all time have to be:

National Lampoon's Vacation
Planes Trains and Automobiles

impossible to just put the funniest movie of all time.. there is way too many films to make that kind of judgement.

that being said a few i remember laughing my butt off uncontrollably are:

i think it was titled "me and him" but i could be wrong where griffin dunne could hear the voice of his private parts... it could of been the mary jane but when i watched this one night i thought i was going to have a heart attack.

something about mary of course one of the funniest.

alot of people tend to think asians don't have a great sense of humor (at least alot of people i talk to) but being an avid asian film enthusiast i find the asian film scene to have some of the more genuine funnier moments in them.. like stephen chow's "king of comedy" there was a scene in there that cracked me up so much... and every film i watch at least gets a chuckle out of me it seems... maybe it's me i don't know.. all stephen chow films generally have at least a few laugh out loud moments.

i remember a little film called "brain donors" with john turturro that i haven't seen in a long time but i remember laughing my butt off when i did see it.

aghh so many going through my mind.. i will just leave it at that for now. hardly any kind of definitive list there is many others.

does this have anything to do with that new laugh and lose weight study?

You beat me to it. This is one of my all-time favorite comedies...and it was made before I was even born.
My favorite characters are the knuckle-headed son ("I'm coming to get you ma"), the tow-truck driver ("that guy's dead"), and the nerds who own the gas station ("We're going to have to kill him"). It always cracks me up
