For me, Airplane! is still the funniest of them all. Others that I find absolutely fall-down hilarious are One, Two, Three, The Producers, Top Secret, Duck Soup, This Is Spinal Tap, Love and Death and Animal House.

The Big Lebowski.
Since this is the DVD Talk thread, I've got to mention that the Director's Commentary to Blade II had me in stiches. Guillermo Del Toro is just too cool. He'll even point out what sucked from his own film.

I recommend "Airplane!" and "All About Eve"

Both Adam Sandler and Bill Murray leave me cold. This puts me in the minority, but it's comfortable there once you get used to it.
No one mentioned the most *amusing* comedy ever made. It's Jacques Tati's "Mr. Hulot's Holiday," finally re-released by Criterion. There's straight-ahead, splat-in-the-face slapstick here, but most of the time Tati is simply observing, with a magical whimsicality, everyday people doing the most ridiculous things. Don't worry about subtitles—the film gets everything over almost without words.

Dumb & Dumber
There's Something About Mary
Office Space
As to some of the people who thought Bad Santa was one of the funniest movies ever, I just didn't think so when I watched that movie... The only part of the movie that got a laugh out of me was when the mom brings the kid over to Billy Bob as he's eating his lunch and he goes off on her about how she could make his life more complicated.
