It's up to you, of course, but there are new areas and story points you are missing out on, and at least one new playable character.

Yes, so the first time through is your "first run". It will then unlock a "second run" for the other character to show their side of the story. If you don't do both runs, you will miss approximately...

You're not wrong, closest I've gotten was in the gun store scenario (in training mode), where the end was in sight but I tried to bum rush it and got chomped by a plant zombie.
