Bowfinger
Death to Smoochy - Edward Norton is great!
Midnight Run (though I'm not sure how underrated it actually is. Everyone I know loves it.)
Joe vs. the Volcano - a lot of heart behind this seemingly low budget flick

Quick Change - IMO, Bill Murray's best

While I agree with a lot of the movies mentioned, comedy is probably the most subjective genre of them all. I have friends for instance, otherwise intelligent people mind you, who won't laugh once throughout Raising Arizona, but think The Waterboy is utterly hilarious.
To the OP, why don't you name some of the comedies you've liked (or the style of comedy you find funny) and then it would be a much easier to task to name other films you might enjoy.

HE Pennypacker....great call on the Russians are Coming, the Russians are coming.
I mean hell, it's got Carl Reiner, Jonathan Winters AND Alan Arkin....
not to mention a greast performance by the late, great Brian Keith.

Yet another vote for Midnight Run. The movie is worth watching for Dennis Farina's character alone.
Similarly aged but also very funny is Running Scared with Billy Crystal and Gregory Hines.
Some other ones that don't get mentioned here much:

The Opposite of Sex
Class
Cousins (good date movie)
Return to Me (good date movie)
Muriel's Wedding
Seems Like Old Times
Next Stop Wonderland
She's the One
And many will disagree, but I personally consider Moonstruck to be hysterically funny. Also, I don't know if it qualifies as under-rated, but Sideways is excellent. See it and definitely listen to the commentary.

The Man Who Knew Too Little starring Bill Murray has been a favorite of mine for a while. It's not a great film, but I really enjoy watching it and laugh so much, everytime.

I agree with a lot of the ones that have been mentioned. Particularly Drop Dead Gorgeous (not as good as the older Smile though), Other People's Money (although I think it's more drama than comedy), and Defending Your Life. I'll add:

House Party 3 Seriously, just ignore the stupid story and enjoy some of the funny scenes (anything w/ Bernie Mac; Ex-Con Catering; Showboat) and hilarious throw-away lines.

Pretty much all comedies are underrated due to them rarely ever winning any Oscars. One of the few comedies that did win an Oscar, Sideways, was kinda lame (wine is not my cup of tea).
Anyway, I agree with the other posters about these movies:
The Great Outdoors
Planes, Trains, and Automobilies
These are two of the best movies I have ever seen.
However, the best movie I have ever seen and the most underrated is:

Planes, Trains, and Automobiles & Midnight Run are two of the funniest movies of all-time, but never seem to get their due by the public at large.
Would National Lampoons Vacation (The Original) count as an underrated movie? Or does it get the respect it deserves, cause that was Chevy Chase in his prime.
