Witchblade Anime Series
Most recently, the
Animated or not; the camera angle, the reasoning, the drama of the situation is captured perfectly. Brings a tear to my eyes even when I think about it.
Spoilered since a couple of members are in the middle of the series.
Reminds me of another great getting out of the building with the baby scene. Hard Boiled! The entire hospital shoot out sequence is a classic! If it's not, it should be...

^^^ As Mifune leans down and just glances over at the slightest movement of Nakadai for his blade, to see if he's feeling froggy. Awesome.
^^^ Some of the most technically correct firearms use I've ever seen in a film in this scene.
The 'car' scene in 'Man On Fire', several scenes from that film are exceptional.
The robbery scene in 'Rififi'.
The robbery scene in 'Le Cercle Rouge', particularly where Montand free hands the shot.
The final confrontation in the street in 'Lawman'.
Any scene from 'Hero', but especially the first scene with Donnie Yen.
Opening scene of 'Aguirre: The Wrath Of God', also the last scene.
Rocky turning down the fight with Apollo, what a scene. Also 'it stinks!' scene.
Opening scene of 'Le Samourai'.
Butterfly scene from 'Mein Liebster Fiend'.
Final scene of 'Infernal Affairs'(Mou Gaan Dou).
Countless scenes from the 'Bourne Trilogy'.
Final minutes/confrontation of 'One False Move'.
Robbery gone bad/shootout in the street scene in 'Heat'. The foley work there was actual gunfire and rounds on cars, some of the best ever in film history.
Taking the beach in 'Troy: Director's Cut'.
Quite a few scenes from 'The Mission', the cross going over the falls, the penitence scene dragging the armor, the final battle.
Frank visits the Italians in 'King Of New York'.

The Right Stuff is full of them, but my favorite is the intercutting between the fan dance and Yaegar's flight near the end of the film.

It's amazing how many people post in a thread based solely on it's title without reading the original post. For me, I think horror movies most typically represent the type of movie the OP is talking about. Jason X has the sleeping bag scene. Ghost Ship has it's opening scene. Deep Blue Sea has Samuel Jackson's speech.
In addition to horror movies with a single scene that pays off, there are quite a few where they're only worth watching for a handful of death scenes. The Final Destination and Saw series come to mind.

I will add the scene in Aliens where Ripley is backing out of the egg chamber as one egg opens, cocks her head, and then lets loose with the flamethrower. Of course, this is just one of many great scenes!

In the form of what the OP described, the Adventures of Pluto Nash and Ernest Scared Stupid fit the bill for me.
Pluto Nash is so terrible but the scene where Mohr tells Murphy he has to swing on electrical cables, dodge fast elevators, and stand on small ledges to get into a casino boss' office. Murphy just stares at him the whole time, ending with a moment of silence and a stare of disbelief, and Murphy responding: "Tell me the other way to get there."
The delivery and it's actual quality at the end of an awful movie made it great.
Ernest fighting the troll with his truck in Scared Stupid: "How 'bout a bumper sandwich, boogerlips!" and then after hitting the troll, he does one of his Ernest-isms, "eh-eww-eww."

The scenes with Luke, Vader and the Emperor in Return of the Jedi are my favorite, especially the last one when Luke refused to turn to the darkside and Vader redeemed himself by saving his son and killing the Emperor. The rest of the movie I never need to see again.
