Lots have been mentioned already but here are a couple of my favorites:

When The Wonders first hear their song playing on the radio in "That Thing You Do!"

I may have missed it but I don't recall anybody mentioning the 3 minutes leading up to, and after the first appearance of the Brachiosaurus in Jurassic Park. I still get goosebumps whenever they show that wide shot and John Williams' score swells and then the dinosaur bellows at the same time. One of The Greatest Moments Ever.

"Show me the money!" from Jerry Maguire

Helicopter rescue scene in Superman: The Movie

The funniest sequence ever - the sequence right in the middle of Slapshot. If you've seen the movie you know what I mean - if not go see it!

Welles destroying his wife's room in Citizen Kane (obvious choice)
The montage of the girl ****ing up the guy's apartment in Chungking Express set to a Mandrin version of "Dreams"
The OD/needle scene in Pulp Fiction
The baked beans scene in Blazing Saddles
The hospital shootout stedi-cam shot in Hard Boiled
The opening motorcycle chase in Akira
The montage of images set to "A Quick One..." in Rushmore(begins with Max putting bees into Bill Murray's hotel room)
Ben Stiller's improvised freak out on the plane at the end of Meet the Parents
The Rolling Stones doing "Sympathy for the Devil" in Gimme Shelter while the crowd freaks out.
ET's goodbye scene in ET
Spacey and James Cromwell in the kitchen in LA Confidential (who saw that one coming?)
The opening pullback on Alex and his droogs in Clockwork Orange
Mifune charging down enemy soldiers on horseback in Hidden Fortress
The cross cutting of Leo getting laid and Tom Hanks doing his laundry in Catch Me If You Can
Batty vs. Deckard at the end of Blade Runner
The "clearing the ghetto" scene set to crazy piano music in Schindler's List

I don't want to reiterate some of the great ones already said but I'll toss out a few others of my own...

1. Race Decoder/Tip Sheet - A Day at the Races - my personal favorite scene from any Marx Brothers movie.
2. Mariachi Trio Shootout - Desperado: Subwoofer does flips everytime the rocket his the limo.
3. The Caravan Ambush - Clear and Present Danger
4. First Organ Scene - The Abominable Dr. Phibes
5. The Voyage of the Gnome - Amelie
6. Business Cards - American Psycho
7. Genesis/Phil Collins Orgy - American Psycho
8. Lawnmower vs. Zombies - Dead Alive
9. The Luggage Sale - Joe vs. the Volcano
10. The Big Win - Let It Ride

Some great ones in here, I'll mention a few more:

1. The entire "ball room" section of Ghostbusters: "Wait wait wait. I've always wanted to do this..."

2. The first real gunfight in Desperado. Those pistols popping out of his sleeve friggin rule.

3. The scene in Gideon's pawn shop in The Crow: "Mr. Gideon, you aren't paying attention."

4. The "meow" scene in Super Troopers.

The end of Leon.
The opening scene from Alien really sets the tone for the rest of the film.
The beginning of The Shining.
The "Processing Station" scene from Aliens.

I've been nodded my head in hearty agreement with many of the choices. For my own:

-'Make 'Em Laugh' sequence in SINGIN' IN THE RAIN. The most amazing preformance I have ever seen.
-The choas at the TV station in the beginning of DAWN OF THE DEAD. I think this is the greatest opening of all time.
-'Under My Thumb' in GIMMIE SHEALTER. The shot where Mick is singing and over his shoulder you see that Hell's Angel guy tripping out. I swear to God it looks like he's turning into a werewolf.
-The industry section of KOYANISQUATSI (however it's spelt)
-The Columbine scene in BOWLING FOR COLUMBINE. As problematic as a lot of that movie is, I've never seen something so torturous and draining on an audience.

From: Second star to the right and straight on 'til morning.
1) The ending of Chaplin's City Lights
2) The breakfast montage in Citizen Kane
3) The final minutes of Casablanca
4) Sunset Blvd. ("All right, Mr. De Mille, I'm ready for my close-up.")
5) STELLAAAAA!!!! from Streetcar Named Desire
6) The final moments on the bus in The Graduate
7) The shower scene - Psycho (probably the most famed of the all)
8) The bloody end of Bonnie and Clyde
9) The baseball sceen in Cuckoo's Nest
10) Ride of the Valkyries from Apocolypse Now

(the hard part of this was to try to find moments/scenes that were 3 minutes or under. Choosing the BEST, however, is a tough one. If we are talking fame I would probably go with Psycho. Really tough to choose)

It's rare I come into a three-page thread like this and my answer has not already been used, but my choice would be the POW camp Russian roulette scene in Deerhunter. That's the most intense scene of any movie ever.
