Lots have been mentioned already but here are a couple of my favorites:
When The Wonders first hear their song playing on the radio in "That Thing You Do!"
I may have missed it but I don't recall anybody mentioning the 3 minutes leading up to, and after the first appearance of the Brachiosaurus in Jurassic Park. I still get goosebumps whenever they show that wide shot and John Williams' score swells and then the dinosaur bellows at the same time. One of The Greatest Moments Ever.

Helicopter rescue scene in Superman: The Movie

I've been nodded my head in hearty agreement with many of the choices. For my own:
-'Make 'Em Laugh' sequence in SINGIN' IN THE RAIN. The most amazing preformance I have ever seen.
-The choas at the TV station in the beginning of DAWN OF THE DEAD. I think this is the greatest opening of all time.
-'Under My Thumb' in GIMMIE SHEALTER. The shot where Mick is singing and over his shoulder you see that Hell's Angel guy tripping out. I swear to God it looks like he's turning into a werewolf.
-The industry section of KOYANISQUATSI (however it's spelt)
-The Columbine scene in BOWLING FOR COLUMBINE. As problematic as a lot of that movie is, I've never seen something so torturous and draining on an audience.
