From what I've seen Paramount wants to make one. I thought I heard Trey and Matt wanted to do one too. I
'm not sure if they are too busy lately since their new movie is coming out soon. It seems the people involved have in interest in a SE.....but nothing has been announced yet.

According to the IGN Rumor list:
South Park: BL&U SE: Paramount is perfectly willing to do it if Matt and Trey would find the time. That's the Paramount line, anyway.

There was a recent interview with Trey and Matt where they were asked about a BLU special edition, and they said that there's no significant deleted material and nothing to really put on a special edition, except a commentary that they weren't that interested in doing. So I wouldn't bet on one any time too soon.
