Lenny and Squiggy's entrances on Laverne & Shirley.
Oh, and Buddy's insults whenever Mel Cooley entered on The Dick Van Dyke Show.

"Say goodnight, Gracie"
"One of these days, Alice, POW...right in the kisser!" & "To the moon, Alice!" (spousal abuse used to be funny!)
"This is the big one! I'm coming, Elizabeth, I'm coming!"

Agent 13 on Get Smart. Always hidden in mailboxes, airport lockers, etc.
The sliding bedroom door on Green Acres
The doorbell on Beverly Hillbillies
Gladys Kravitz seeing something while peeking through the window on Bewitched
The signs on the batcave equipment on Batman
The cannon on F-Troop
Hogan's Heroes-when Schultz walks in to barracks and sees people who aren't supposed to be there, then turns around and leaves without saying anything pretending he didn't see it.

"Hi! It's me, Stan!" -- Golden Girls
The random insertion of rock lyrics into dialogue in St. Elsewhere. For example, Dr. Fiscus's reaction to the hot blonde Crane nurses. "You Crane girls really knock me out..."

Another Get Smart one
When somebody gets shot or something, and is laying on the ground dying, someone kneels down over the body. The person dying gasps like he has something to say. The person kneeling will lean down, ear to mouth, to hear the dying words. Someone will ask, "What did he say?" The person who knelt down will always reply, "He said, get your knee off my chest".

One that popped up occasionally in the Three Stooges shorts...
Curly or Larry: I can't see! I can't see!
Moe: What's the matter?
Curly or Larry: My eyes are closed!

I use this one myself in every day conversations. Those who know what I'm refering to find it funny.
A couple that immediately spring to mind are Seinfeld running gags:
Kramer's jacket
Kramer constantly screwing Susan over. (He vomited on her, burned her cabin down, stole her lesbian lover)
George refering to himself as "George". "George is getting upset!"

Thought of another one: Shawn's "I've heard it both ways" on Psych. I also enjoy the fake names he comes up with for Gus.

Which means we should also include never seeing Maris (sp?) on Fraiser. To me Maris/Fraiser was a bigger gag than Vera/Cheers. Cheers was centered around the bar - not a lot of opportunity to see the personal lives of these characters anyways.
Fraiser was centered around the family members (whether they were in Fraiser's apartment or at some social function) and were almost always together. It was always funny to hear from Niles about the unfortunate recent event in Maris' life that would stop her from joining the family.
