When does Amazon deduct the coupon amount on multiple items?

When does Amazon deduct the coupon amount on an order for two items? Will they take it off the first shipped item or on the last shipped item?

The first shipped item. So if you want, you can have an item shipped with the coupon amount deducted and then cancel the order.
