Tough to say but it would probably be Trading Places. It just makes me laugh and puts me in a good mood every time I watch it. Animal House does the same. Along with Vacation.

+1 Something about this one is hilarious and utterly re-watchable. It has a completely uneven mix of deadpan subtlety and over-the-top absurdity but somehow it works.

Whenever I was depressed, I used to put in Monty Python and the Holy Grail. Kind of lost its luster, but for a while, it was a great antidepressant for a while.
