Favorite running joke
I'm watching News Radio on DVD, and I've found there is one surefire way to get me to laugh: have a character turn to Joe and say "Wait -- your last name is Garrelli?" It never fails to crack me up.

In Family Guy, whenever Peter gets kicked or hit in the shins and his reaction to it never fails to crack me up.

Robot Chicken
- Gag credits that change each time for Sarah Michelle Gellar, Mila Kunis, and a random Chicky Robo employee.

I just watched the episode where Charlie put the bar down as the prize in the dance contest because he had "pride" in the bar. 
The running joke I really like about Always Sunny is the way they cut to the title of the episode.
So many good running gags on that show. My favorite has to be "It's an illusion, Michael. A trick is something a whore does for money." Either that or the chicken impersonations ("Has anyone in this family ever seen a chicken?")
