This can be the voice acting, the cutscenes, the gameplay itself, anything.

My current 2 choices are:

Giants: Citizen Kabuto (the Mecs and Smarties are just too funny -- "for I'm an old man and me balls are saggy...") "What, those smarties are real.. and just yesterday i was throwing rocks at them.")

and

MDK2 "Come on boy, strap yourself to a torpedo while I figure out how to aim...I love surplus." "Hello Earth, who's your daddy? why yes, I am."
Also, those cone headed farting aliens. That's really funny. (you gotta see it to believe it)

The voice acting in both these games totally rocks, and the gestures are hilarious too. I know at least for MDK2 that they actually hired a comedy troupe to do a bunch of the voices and it paid off IMO. If only more people bought this game -- it's a ton of fun too. (and only $10 at compuexpert.com)

Hee, hee; Custer's revenge is pretty funny. Very black humor, mind you, but still funny.

Conker's Bad Fur Day might be on this list. Haven't played it yet, though.

Dungeon Keeper 2 Delightfully wicked and funny - hearing the narration is worth the price of the game itself.

Parappa the Rapper. To rap your way to the front of the bathroom line because you ate a seafood cake is hilarious!
