He still made occasional appearances, and even though I really liked the addition of Ben Browder and Claudia Black, I think Jack O'Neill was the best character on the show and I was sad to see him leave.

Every character that left M*A*S*H, but since no one has said him yet, I will say Radar, the only actor to reprise his role in the film in the series.
Half the cast of the Practice in the final season, although the Spader intro/Boston Legal set up was awesome.
Rhoda & Phyllis the Mary Tyler Moore Show..
Mr. Roper Three's Company... Although I really liked RF...

Anthony Stewart Head (Giles) on Buffy. Once he went from regular to occasional guest star, I thought the show lost a lot of its emotional and grounded core. There were MANY times during the last two seasons where it just seemed like a bunch of kids screwing around (sometimes literally) and doing stupid things without anyone to tell them no.
Show definitely went downhill when he left.

The Newsradio and Coupling examples cited are perfect ones. I join them.
The Shield did not feel the same after Kenny Johnson/Lemonhead was gone, but that might have to do with the circumstances more than the character himself.
I would think the loss of Nancy Marchand as Tony's mother on The Sopranos would have also been mentioned by now, but I don't see it.
I think the loss of Adebisi (Adewale Akinnuoye-Agbaje) on Oz was a significant one, too.

I liked Pulaski. Her lack of respect for Data created a much-needed bit of tension that helped the show find its footing if you ask me. I don't think I missed her when she was gone, though. She served her purpose. I think Diana Muldaur went on to create the same tension among the attorneys at McKenzie, Brackman, Chaney and Kuzak. Her demise on LA Law is one of the greatest moments in TV history if you ask me.
I do think I missed Gates McFadden during that season, though. I think she qualifies for this thread.
Church as Lowell on Wings is a great call.
I disagree with Tasha Yar on TNG and Shelly Long on Cheers. I didn't watch Cheers from the beginning, so that might explain the great amount of dislike I have for Long's character. I also watched M*A*S*H somewhat backwards, which is why I probably prefer Winchester to Burns, slightly prefer Potter to Blake, and find Hunnicut to be pretty much just as good as Trapper. I don't hate any of the originals on M*A*S*H like I hate Long's Diane from Cheers, though. As far as Yar goes, I think her departure led to much more interesting stories for Worf. That was a good thing. He was a far stronger character than she likely ever would have been.
I think the only Star Trek departure that ever really bugged me was Jadzia Dax, but even it was designed in a way that propelled the story forward in a satisfying way (plus, it's not like they had a choice). That Terry Farrell really did it for me, though.

Rob Morrow (Dr. Joel Fleischman) on Northern Exposure. The show took a nosedive in quality and only lasted for eight more episodes before they pulled the plug. He probably had one of the best exits on tv imho.

Rob Morrow was the glue that held "Northern Exposure" together. I know it was an ensemble show, but his character always seemed to be the one thing that held it together.
That and the writing for the last season was off compared to the previous seasons.

Surprised no one has mentioned Delta Burke & Jean Smart leaving "Designing Women" after season 5. Going from what is arguably your peak during the prior three seasons to the pathetic shell the show became as it limped to cancellation is still rather astonishing.
