I didn't think it was really a shot at Oprah as much as "Look how stupid Jenna is/Liz can't act for shit".

In addition to showcasing how stupid Jenna is because she doesn't know the difference between Slingblade and Oprah, it's also funny because one of the basic rules of Improv is that you never respond to your partner by simply contradicting him. All it does is kill the scene. So Jenna's not only stupid, she's also a bad improver.
