I assume you're asking about Buffy the Vampire Slayer. Plotwise, I don't think I have to tell you why they changed principals. As far as behind-the-scenes reasons, I don't think there were any. Joss Whedon has never been afraid to kill off characters.
That's one of the things that made me realize how different this show was from anything else on TV. Jesse seemed like he was gonna be one of key members of the Scooby Gang, and Joss turned him into a vampire and staked him by the end of the second episode of the series.
