Of course "never heard of" is relative, but here's a batch of folks that I listen to all the time that sadly aren't too well know:

Alejandro Escovedo
Jon Langford

Well, I've heard of all four mentioned (and love everyone listed by 12thmonkey). My favorite lesser known musician is Robert Randolph...an incredible pedal steel guitarist. I don't think he'll remain unknown for long.

I'm not sure that anyone who has had a song featured in a Volkswagon ad can really be an unknown, but I agree that Nick Drake is time well spent. Pink Moon will change your life.

As will anything from
Ali Farka Toure - a terrific African guitar player; the delta blues by way of Mali
The Campbell Brothers - sacred steel, gospel music with weeping and wailing steel guitars. Wow.

12thmonkey... I love Alejandro Escovedo. Excellent guy. I've also seen the Mekons in concert a few times and love their work.

I can't think of the greatest unknown musician ever, but some good unknowns would be Ron Sexsmith and Freedy Johnston. Of course, that term "unknown" might not work with radical music fans.

I'm going to have to cast my vote for The Handsome Family. They're one of the funniest bands I've ever heard, and hardly anyone knows who they are!
