Best Film Songs
(I'm aware I am double-posting after watching My Fair Lady)
I hated musicals when I was younger, but now I find an appeal to them. It's funny how many of their songs I knew before I'd actually seen them.
What are the best musical (or music-centric) moments in film, where the characters are involved in the song?
The three that come immediately to mind:
Singin' In the Rain - Singin' In the Rain
Once - Falling Slowly
Topsy-Turvy - Three Little Maids

I love all of the responses so far, except for Oldman.....that was a POS movie that defiled the punk rock movement. It was The Theory of Everything for people that wanted to be "in the know" for everyone that was not "in the know". Fucking hate that movie.

I'd have to put various numbers from WEST SIDE STORY at the top of the list:
The Jets Song
Tonight
Gee, Officer Krupke
I Like to Be in America
Cool
If you forced me to pick something more recent, I'd go with the "Colors of the Wind" number that Pocahontas sings in Disney's POCAHONTAS.
I'd also second Mabuse's picks from a few posts up. They perfectly illustrate the OP's dictum of the characters being involved in the song:

The thread title seems to be asking for best songs that originated in films, but the OP then asks for best musical performances in film.
For the question I thought I was going to answer, I would say stuff like Unchained Melody (from the movie classic Unchained) and Charlie Chaplin's Smile.
For the actual question, I'd counter with
The Man That Got Away - A Star Is Born (1954)
Over the Rainbow - The Wizard of Oz
Cheek to Cheek - Top Hat
I Got Rhythm - An American in Paris
And to piggyback on the John Carney selections already named:
Begin Again - Lost Stars, Tell Me if You Wanna Go Home, Coming Up Roses
