Just thought of a few more:

Sean Astin - Rudy/Samwise Gamgee
Elijah Wood - Frodo Baggins
Mandy Patinkin - Inigo Montoya
Cary Elwes - Westley (From The Princess Bride)/Robin Hood (from Men in Tights)

And who could forget as one of the greatest cinematic trios -
Harry Shearer, Michael McKean, Christopher Guest - Spinal Tap

Keanu will always be Neo to me too. Hugh Jackman as Wolverine is a nice one. Kevin Spacey as Verbal Kint.

Perhaps Bill Murray in Rushmore? I don't quite understand the question, is this asking for roles which people remember actors by?

I don't think a lot of these responses really are answering the question. I think the idea is that the actor is generally thought of in that role, regardless of what he did. I don't think someone like Tom Hanks or Arnold would even fall into this category as their status as a star outweighs any particular role. From there I think we're just naming our favorite roles of certain actors. No, a good example would be Dustin Diamond as Screech. Who knows what else he's done, he's Screech for life. But that's TV.

I'd personally go with Matthew Broderick. I'm not even sure what else he's been in. Everytime I see him I think Ferris Bueller. Hell, I have a hard time remembering what else he did...cause...he's Ferris Bueller.

I almost listed that role, but passed because it was TV.

If TV counts (although they have done movies of the first two shows), I guess you could list most of the Star Trek actors. Exceptions would be Patrick Stewart (because of X-Men), and some more established actors like Scott Bakula and Kate Mulgrew, who had done some film work and were somewhat known before joining the casts of their particular series.

I don't know, but when I think of Keanu, I think Whoa, Dude! - Ted, from Bill & Ted.

I only mentioned Jack Traven cuz that's the role that put him on the map and in the process, in sight of other big time directors/producers.

Christopher Lloyd - Doc Brown
Crispin Glover - George McFly
Alan Ruck - Cameron Frye
Jeffery Jones - Ed Rooney

I've seen enough convincing stuff from Matthew Broderick to be able to identify him with other rules, but I guess first and foremost, he's always Ferris.

Just thought of one that I'm surprised hasn't been mentioned yet...

Mark Hamill as Luke Skywalker

Now that was clearly a roll of a lifetime for him.
