Wow, what a tough one, since there are so many great comedies (many of them mentioned above). To help my decision, I used the following criteria (leave it to me to attempt to make this scientific somehow):
1. It had to make me laugh out loud, not just smile or chuckle;
2. It has to hold up to repeat viewings (be funny each and every time I watch it);
3. It has to be something that I want to watch multiple times, even if I happen to catch the edited version on TV while flipping through channels;
4. It has to be quoteable; and
5. It has to be universally funny (that is, other people I watch it with have to also "get" the humor - there can't be too many inside jokes or humor so skewed that I'm the only one in the group who laughs).
With that being said, it was a stretch to even limit my list to four, but here goes:
1. Caddyshack
"It's in the hole!"
2. Airplane!
"And don't call me Shirley"
3a. (tie) Animal House
"Toga! Toga! Toga!"
3b. (tie) Planes, Trains, and Automobiles
"Those aren't pillows"

Wow some movies here that I disagree with strongly. Personally the South Park movie didn't make me laugh a single time, I hated it and I love the tv show. Just goes to show you how folks have different tastes.
