...that Dirty Harry's famous quote from Sudden Impact--

I, too, haven't yet looked at the list (thanks for the link). A few that come to mind off the top of my head, though...
Citizen Kane's "Rosebud"
King Kong's "It was beauty killed the beast"
Heston's famous quote from Soylent Green (don't want to spoil for the two people on this board who haven't seen it )
Heston (again!?) from Planet of the Apes - either the "Get you hands off me..." line or the revelation on the beach (again, don't want to spoil).
The Graduate's "Mrs. Robinson, you're trying to seduce me" line.
Plus any of several good ones from Casablanca.

I just got done watching The Untouchables for the first time and am a bit surprised that the below quote did not even make into the Top 400.

AFI seems to have a thing for Wizard of Oz so I'm guessing "we're not in kansas anymore" or "there's no place like home" tops the list. One of the quotes from Casablanca will come in second or third though I was disappointed that "I'm shocked, shocked to find gambling going on in this establishment" was not one of the nominees.
Big Dave, I'm with you on The Untouchables quote but I have a Chicago bias as well. I'm hoping "you're gonna need a bigger boat" makes it into the top 5.
