Mine had to have been during Happy Gilmore when Chubb showed his hand that got bit off by the alligator. I don't know if it was because it was super hilarious, or I was hopped up on Ephedrine. Probably a combination of both.

Wasn't me, but I'll never forget a friend losing it watching Dumb and Dumber. He lost it so much during the snowball scene, that he fell off the couch doubled over crying with laughter.

The whole In The Loop movie had me rolling. Every scene with malcolm and the dinner scene where he tries to defend his cheating by claiming he did it to stop the war. Also Four Lions was absolutely hilarious.

All the Tony Clifton scenes during Man on the Moon, but especially the one on the set of Taxi.
In Dodgeball, after Missi Pyle as the Russian brute destroys the guy in the bar with a ball, Justin Long whispers, "I think that guy might really be dead!"
Gene Wilder and Richard Pryor reacting to the monster Grossberger in their cell in Stir Crazy.
The Little Seymour scene from Uptown Saturday Night.
Any one of a dozen classic Abbott & Costello bits, but especially the drill scene from Buck Privates or the Susquehanna Hat Company bit from In Society.

The one I remember most vividly is the "You're going the wrong way!" scene from Planes, Trains and Automobiles. Such a great slow build. About 1:20 in the clip is where I started to lose it - tears, snorts, doubling over - everything.

Years ago (stoned, naturally ) watching the teen comedy "Hollywood Knights". There's a scene with a nerdy guy and his mother in a car singing songs from "Fiddler on the Roof". That set me off so hard that I actually cried. LOL

I don't know why (it's only sort of funny now), but the hardest I ever laughed at a theater actually came the first time I saw the trailer for Baseketball. When Bob Costas yells out at the end, "You're excited? Feel these nipples!" I remember literally falling on the floor with tears falling down my face I was laughing so hard.

I saw Monty Python and the Holy Grail in a packed theater. Everyone was in hysterics by the time a moose once bit my sister. We never stopped.
The only thing close to it was the Kentucky Fried Movie / Groove Tube double feature that all the guys from my dormitory floor saw together at a beer theater. There wasn't a dry eye in the house during Catholic High School Girls in Trouble.

The Mrs Doubtfire telling off the mugger bit is a great one.
Two parts from Dumb and Dumber:
- Harry is choking Llyod who screams out "Harry... your hands are freezing!"
- The lady is reunited with her husband and suddenly Llyod becomes enraged and starts shooting at him but he wakes up.
Transformers:
- where the soldier tries to make a phone call and rushes out to get a credit card. Well, it was very funny the first time.
Full Metal Jacket: "Wipe that smile off your face!"

When I was about 11 years old seeing Home Alone for the first time I remember laughing hysterically at Daniel Stern's scream when he has the tarantula put on his face.
