One that comes to mind is Monty Python's Life of Brian, in particular the "Biggus Dickus" scene, where the centurions are trying not to laugh as Terry Jones "Bigggggggggusssss..................Dickus" at him. I thought I was gonna lose it.
Then in South Park, the opening "Uncle Fucka" song. I was laughing so hard at that, and finally caught my breath as the song was ending. And as they're holding out the final note, Terrance (or maybe Philip?) just says "Suck my balls" for no reason at all. I nearly busted a gut literally on that one.

The second hardest I remember laughing at a movie was Airplane. The hardest I remember laughing at a movie was the preview for Airplane a few weeks before it came out. It wasn't like now where you know everything coming out well in advance, and I'd never even heard of it. It wasn't just me, practically everyone in the place was laughing hysterically. I know it was in Westwood, but I can't remember what movie I was actually seeing.
Monty Python and the Holy Grail was damn high on the laugh scale too, especially since it was the first time I'd seen any Monty Python.

There's Something About Mary - When Ben Stiller goes to Mary's house to meet her for the Prom and touches Warren's ear by pulling the baseball out of thin air. Warren beats the shit out of him, and for any Wrestling fan he 'racks' Ben Stiller (A Lex Lugor move).
Slap Shot - The first shift of the Hansen's where they totally demolish the other team, and then the one of the Hansen brothers shoots the puck at the Organ Player and hits him in the head!
Dumb & Dumber - Where Lloyd Christmas is driving at night and has a dream that he is married to Mary Swanson, which shows all these different scenes (He gets beat up by the Asian Cook and then rips his heart out in an ode to those old Kung Fu Movies, lights a fart with a zip lighter, etc.)
I'm sure there will be more that comes to mind as I will post them later....

Welcome To Collinwood – So many scenes in this one. But the ones that get me into fits of laughter is Toto and his pants, and then Toto and his underpants. OH MAN!!! I fall apart each time he is on screen afterwards with the damned jacket. Also the wall drilling scene.
Gran Torino – Walt talking to Thao, being his obnoxious self and not getting any names right. I was cracking up with “Click-Clack” and “Ding-Dong” but by the time he said “Yum Yum” I completely lost it.

I don't think I've ever seen a theater explode into laughter so hard as during the "I just shot Marvin in the face" scene in Pulp Fiction. I'm not sure what that says about us as a society.

At the theaters, some that come to mind are Borat, The Waterboy, You Don't Mess With the Zohan, Dumb and Dumber, There's Something About Mary, and Wayne's World.
A lot of others have been mentioned that I saw at home. The last movie that had me laughing almost the whole time was This Is The End. I just don't make it a point to see a lot of comedies at the theaters. It's nice to be in a crowd totally get into it, but I hate paying night-time prices for movies anymore.

I also have a weak spot for Steve Carell's brand of comedy, so both the possessed anchorman scene in Bruce Almighty and pretty much all of The 40 Year Old Virgin get me rolling.

I forgot about Team America. I was laughing myself silly in an empty theater on opening weekend. We really need a stateside BD.
I'll also add Ace Ventura: When Nature Calls. I saw it in theaters but I was too young to really remember what I thought was funny. After revisiting it for the first time in years awhile back I absolutely lost it during the rhino scene.
The first Ace Ventura I always liked as a kid but I clearly didn't understand many of the jokes growing up. When I watched that for the first time in years I was on the floor. I still think it's Jim Carrey's best movie.

It's been mentioned a few times already, but that bar scene in Team America. I think I missed the following 5 minutes of the film because I was laughing so hard. I can't remember who I saw it with, but their reaction was identical.

Yes, FMJ had me rolling. Wonderfully made film, you know that it is horrendous what they are going through but it is hilarious at the same time.
