The two I thought of right off the bat were Sleepers and Now and Then, just because both of those have big pretty big ensemble casts with many recognizable faces.

Ken Watanabe and Liam Neeson as "Ras Al Ghul" in Batman Begins

Wil Wheaton and Richard Dreyfuss as "Gordie" in Stand By Me.

LeVar Burton and John Amos as "Kunta Kinte/Toby" in Roots.

I don't know if in-movie films count or not. Technically, the real actor is playing an actor within the in-movie film, not really the same character.

I thought of it, but since the only reason Quinto is famous is because of Star Trek, I don't think it really counts (unless he was known for something that I'm not aware of).
