You know what I'm talking about - the villian is standing there, thinking he's got the upper hand. The hero balls his fist, rears back and lays him out with one triumphant blow!
I was watching Wedding Crashers again this weekend and, I have to say, Vince Vaughn knocking Bradley Cooper on his ass not only looks extremely painful, but is an extremely satisfying moment all around.
This year: Optimus Prime's close up slo mo uppercut causing Bonecrusher's optical sensors to bug out and his face to distort.

James Cameron's The Abyss:
During the tense fight between Navy Seal Coffey (Biehn) & oil-rig driller Bud (Harris) where it was quite obvious who's gonna get his ass kicked, Bud's friend sneaks up behind Coffey and gives him a wallop of a punch.
I've always loved the punch that introduces Harrison Ford in "Last Crusade":
Young Indy gives Cross of Coranado back to tomb raider.
"You lost today kid, but that doesn't mean you have to like it".
Tomb raider puts hat on Young Indy as he lowers his head.
Head raised up to reveal Harrison Ford Indy in 1938.
Harrison smiles at camera.
THWAP! Gets slugged across face.

Pictures are really the only accurate way to show it:

I prefer the fight with the German airplane mechanic in the first Raiders. That first punch he takes that puts Indy on his ass.

Yep. I haven't watched it, but based on that "highlight clip" it must be pretty freaking awful.

It is by and large one of the worst remakes ever. I love the original, the remake didn't even scratch the surface.
That said, it is hillariously bad, I mean, laugh out loud awful.

George McFly
Morgan Freeman's final punch to Shawrelle Berry (black punk at gym), in Million Dollar Baby.
First punch I thought of. Every time I watch the movie I always think the kid is going to knock Jason off the roof. Just 1 or 2 more punches...such a shame. ;)

I'd never seen that movie, but that dude's problem started when he punched a guy who was wearing a hockey mask.

All the punches in the Indiana Jones movies are pretty good. You can tell a punch is from an Indy film just by listening to it...They freakin' echo!

I was thinking the same but not so much the punch itself (as it's a quick jab), but Indy's reaction is great -- get hit, pause, wobbly legs, fall on ass

Most of the good dramatic or action-packed ones have been mentioned, but I've always been fond of Kelly Preston's shot at Tom Cruise in Jerry Maguire.

I know it's not a movie but a great punch nonetheless.
On Entourage (I just caught up with the series on DVD):
In the Vegas episode. Drama punches Seth Green's friend at the end. You don't see it coming and it is a big payoff to what was building in the entire episode. I cheered like I was watching a sporting event. It was great The greatest TV punch ever I guess. That is when I knew I loved the show.

I second the one in Raging Bull. The artistry in that sequence still blows me away every time I repeat-view that movie.

Billy Zane's punch in Demon Knight, where he puts it straight thru the Sheriff's head.
Crank. When Chev Chelios dodged and the Samoan guy drove his severed wrist bones (ulna + radius) into the grimey asphalt. I was rolling.

The McFly punch in Back to the Future is definitely the best. Because it's not expected. When you watch boxing, you kinda do expect punching to be involved.
