I thought Eulogy was really good and under-the-radar, but maybe I just missed it when it came out.

Romy and Michelle's High School Reunion. It's pure genius, and I have no idea why it never caught on.

Planes, Trains & Automobiles is my all-time favorite comedy. It's well-known, but never really was a bit "hit" (in the theaters or on home video).

Still Crazy
Blast From the Past
Defending Your Life
The Out-of-Towners (original only)

Slackers is a pretty bad movie but Jason Schwartzman comes pretty correct. Worth a rental.

I think "Ali G Indahouse" is hilarious (and more fun than "Borat") but most reviewers disagree.

The con makes absolutely no sense whatsoever but I still found the 2004 version of The Big Bounce to be very funny. It is not strictly a comedy but I think Wasabi is pretty funny.

Also, I liked Bubble Boy (so ridiculous) and The Cable Guy (vastly underrated).

Hmm, I think 10 ways to lose a guy and witherspoon's blonde films are last time I laughed at comedy. Actually I was thinking other day about this, since 80s, death of John Candy and such this genre has changed alot.

I can't understand the appeal to this movie. I loved the Jon Lovitz bit, but that was about it. Trying to turn child molestation and rape into a lighthearted idea, I don't know. It just wasn't for me.

I agree with Midnight Run and Bowfinger.

Also:

Back to School (probably the best Rodney Dangerfield movie, aside from Caddyshack)
The Great Outdoors
Grosse Pointe Blank
The Road to Wellville (not the greatest movie, but definitely a different comedy than you're used to seeing)
Throw Momma From the Train

These have been said already, but I agree with:

Midnight Run (it gets better everytime I see it).
Throw Momma From the Train (different, and better, than I thought it would be).
Bulworth (Watched it in my college philosophy class. I didn't really care for the ending though)

Not previously mentioned:
